At the inlet i want to give a boundary condition which is the d(uds 0)/dt. so for this which macro to use?

I have tried DEFINE_INIT and DEFINE_PROFILE.

Both gives me an error during initialization of the solution.

I think it is simple there many macros in udf that you can use to write you program, you'd better see the help file for details.
