If you have specified a Velocity-inlet boundary condition it would be good to use it ('Compute from' drop-down list in the Solve-Initialize... menu) for initializing the domain.

But generaly- initializing depends on the BC you've set.

it is simple,

As first step run your solution as steady state.

secondly change your solution type to unsteady state and solve it. fluent will automatically take your steady state results as initial condition for your unsteady case.

beware, never to intilize your solution when to solve unsteady case because it will over write your steady state results
