Best Way to Capture Boundary Layer Thickness
Can some body tell me what is the best way to capture the boundary layer thickness of an fluid flow in any CFD solver.

I don't know if I got your question right...but I assume you need to properly calculate your Ywall (height of first cell) based on the deisred Y+. Rule of thumb: 9-15 cells should be within your boundary layer in order to have good representation.
You can check your result by checking the contour plots of Y+ distribution and addapting your grid accordingly.
