Interface is one which having two sharing surfaces, in interface option it will combine two surface into one.. We can see face deleted in execution window..
If in yur case two fluids are one above...

After cell-zone conditions> Mesh Interfaces if its Fluent 13/14.. older you can get by menu bar.. Define> Mesh Interfaces..
Bt you wont be able to access it unless you define it while meshing.....

Name the faces of Interfaces while meshing,,
In Fluent interface > create interface >Select source face and target faces..
If you are working on heat transfer, you can opt for Coupled wall.....
