Is it possible to initialize the domain by assigning angular velocity?

Yes, I think so. You can set the initial condition as a field function in the initial conditions of domain.

Yes. Use a cylindrical coordinate system. Align the z-axis with the rotation center. Specify the initial condition against that coordinate system.
